CBSE Biology Investigatory Project Class 12
In the Bio Investigatory project Class 12, the experiments comprise a defined procedure and a number of steps to be followed by the scientific investigators to answer the questions about the natural world. It also includes making an observation, determining a hypothesis, and arriving at an inference or conclusion. - Concentration of Salt in Plant Cell Experiment
Objective: The main purpose behind holding this experiment on a plant cell is to see how the different salt concentrations affect the plant cell’s cytoplasm when exposed to the salt.
- Graduated cylinders
- Red onion

- Table salt
- Small knife
- Paper towels
- Distilled water
- Paper, pencil
- A spoon
- Gather all the material that is needed for a plant cell science experiment and put on the safety goggles.
- Cut onions into two wedge-shaped pieces.
- Use an eyedropper to drop the distilled water in the centre of the microscope slide.
- With the tweezers, peel the thin layer of the onion wedge and put it in the centre of the microscope slide.
- Add a drop of distilled water and one drop of the stain like iodine or eosin on the onion tissue placed on the slide.
- Allow the air bubbles to escape by carefully lowering the glass slipcover at an angle above the stained tissue.
- Examine the slide under a compound microscope at a magnification of 100X.
- Draw a sketch of the image and identify the observed structures.
- Prepare a 5 percent solution of salt by adding 5 gms of salt per 100 ml of distilled water in a cylinder. Gently shake th mixture until the salt gets dissolved. Prepare the 10 percent of the solution by adding about 10 gms of salt per 100 ml of the distilled water in another cylinder.
- Add 5 percent of the solution with a dropper to one part of the slide. The 5 percent solution should be mixed well with the liquid which is already placed on the onion tissue slide.
- Now glide the triangular end of the paper towel under the opposite end of the cover slip. This will mix the liquid on the slide. Add more drops of the 5% solution to complete the introduction of the new solution.
- Repeat the process in steps 7 and 8.
- Use the dropper again to add a few drops of 10% solution to the 5% solution.
- Glide the triangle-shaped part of the paper towel on the opposite end of the cover slip. Use some additional drops of about 10% solution when needed for completion of the new solution.
- Repeat steps 7 and 8 for your plant cell science project.
Conclusion: After recording the data, students will learn how different salt concentrations can affect the cytoplasm as well as the growth of the plant.
2) Haemoglobin Test
Objective: The Haemoglobin test experiment helps students get an idea of the increase in the generation of haemoglobin in a species like Daphnia Magna that responds to the decrease in oxygen concentrations level. It also aims to know the role of haemoglobin in the body and also the effects of an increase in its level on the animal and human body.
- Culture Kit for Daphnia Magna
- A microscope or a magnifying glass
- A digital camera
- Coloured pencils
- Petroleum jelly
- Plastic wrap
- Take out the culture kit for Daphnia Magna and follow the instructions to set up and establish the culture. Use the microscope or a magnifying glass to be able to see the small details of their bodies. Take a picture of the initial culture with the digital camera and then draw a sketch of the individual Daphnia. students can also use a bit of petroleum jelly in order to hold the individual Daphnia for the purpose of further observation. Students should ensure that they colour the sketch in an accurate manner with the help of different coloured pencils.
- Arrive at your own hypothesis. What will happen to Daphnia if the concentration of oxygen in the environment goes down and why? Feed the Daphnia in order to prepare it for the low oxygen level experiment.
- Cover the surface of water carefully in the aquarium with plastic wrap. It will prevent the gases from leaving or entering the surface of the water. When the Daphnia uses oxygen and produces carbon dioxide, the dissolved oxygen concentrations in the water will drop. Observe the Daphnia for the next two to three weeks. every day, take a picture of the entire population and take notes of the changes happening. Students need to pay keen attention to the colour of the Daphnia. As they are also called the transparent species, the change in their colour will be a direct outcome of the change in their blood and the haemoglobin synthesis.
- After about 2 to 3 weeks, take off the plastic wrap, take a picture of the population, and prepare another sketch of the individual Daphnia. Colour the sketch properly in order to represent the accurate changes that took place. Feed the culture and then allow it to get used to the normal conditions without the use of the plastic wrap. Take down notes on it every day and also the pictures. After nearly a week, if you notice that they are getting back to normal, sketch the other daphnia.
- State your inference based on your hypothesis.
Conclusion: Daphnia Magna, the small crustaceans living in freshwater lakes, ponds and streams, feed on the algae and are known to be very sensitive to bad water conditions. These are also used by scientists to determine the quality of water. Haemoglobin is a protein which is present in our blood and carries oxygen from the lungs to cells of various living organisms. The experiment changes the oxygen levels of the water where Daphnia are placed to evaluate the production of haemoglobin in these creatures.
